1. In the videos, John indicates that the silk screening around the edges of the plates are there to separate them ever so slightly. Further, he mentions that the sample must not be a solid or something that will change that separation. So keep your sample inside the border.
The sample doesn't need to touch anything, just put a drop or two of your sample in the center of the plate and set the other on top. It's nice and simple.

2. The instructions say to cover the “E” so that is what I do. To not completely cover means it leaves room for static noise of empty space. A better scan will fill the “E”.
